The present study investigates a green route for the synthesis of copper nanoparticles (CuNPs) using pomegranate (Punica granatum L.) juice as a natural reducing, stabilizing and capping medium, and evaluates the antimicrobial activity of the synthesized nanoparticles. Pomegranate juice contains phytochemicals including flavonoids, proteins, tannins, phenols and terpenoids that can participate in the reduction and stabilization of metal nanoparticles. CuNPs were prepared by adding diluted pomegranate juice to a heated copper oxide precursor under stirring. Two juice volumes, 3 mL and 6 mL, were investigated. The products were characterized by UV–Visible spectroscopy, X-ray diffraction (XRD), scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDX). UV–Visible spectra showed absorption features at 220 and 232.5 nm. XRD analysis indicated crystalline Cu-containing nanoparticles, with an average crystallite size of 46.30 nm according to the reported Debye–Scherrer calculation. SEM images showed irregular particles and agglomerated/flaky structures in the approximate 50–100 nm range. EDX analysis confirmed copper as the major element in the samples. Antimicrobial activity was assessed by the agar well-diffusion method against Pseudomonas, Proteus, Staphylococcus and Bacillus, producing reported inhibition zones of 23, 17, 19 and 22 mm, respectively. The findings demonstrate the potential of pomegranate juice as a simple and low-cost biological medium for the preparation of copper nanoparticles with antimicrobial activity.
KEYWORDS
In this study , we successfully synthesized copper nanoparticles(CuNPs) using one-step ,eco-friendly and cost effective green synthesis method. The bio-components present in punica granatum (pomegranate ) juice served as excellent reducing and stabilisng agent during the synthesis process. The UV-Vis and XRD analysis confirmed the high purity and cubic crystal structure of the CUNPs phase, with a crystalline size of 46.39 nm and it is in agreement with SEM morphology. The green-syntheised CuNps reveal cubic crystal clusters with different lattice constants. The biosynthesized route is more important than the chemically because it offers a new means for researchers to produce cost-effective ,highly active sites with high purity, non-toxic,eco-friendly and have low energy consumption. These efforts will definitely facilitate the translation of synthesis of CuNPs from the laboaratory scale to large –scale industrial production with the goal of benefitting human health and for sustain the protection of environment with chemicals. The absorption phenomenon in copper nanoparticles, due to surface plasmon resonance is found to be, prominent at about 220 and 232.5 nm .The XRD indicated the presence of CuNPs with significant peaks at [111]. EDX shows the elemental composition is above 80% in the sample. Through antimicrobial activity CuNPs have explored the long term mechanism in antimicrobial efficacy which would open up new avenues in the field of nanomedicine.
